Understanding Diospyros rumphii Bakh from North Sulawesi through Long-read sequences analysis using MinION Oxford Nanopore Technologies

Abstract Diospyros rumphii (Ebony) is a high-value timber species native to Indonesia, which listed as Data Deficient by the International Union for Conservation of Nature (IUCN) Red List Threatened Species. Therefore, additional data enrich knowledge on distribution, ecology, and genetics this urgently needed. Furthermore, study aimed generate whole-genome sequencing data, including low-coverage D. using MinION Oxford Nanopore Technologies (ONT). The sequence generated from leaf cambium genomic DNA was assembled result in draft chloroplast genome constructed phylogenetic tree based partial nucleotide sequences rbc L gene. results showed that 3.25 Gb long-read with size 150,578 bp. Genetic relationship analysis gene positioned one clade oblonga , has bootstrap value 87% suggesting can be considered recommended molecular marker genetic future.
